末端脫氧核苷酸轉(zhuǎn)移酶（Terminal Deoxynucleotidyl Transferase，TdT），是一種不依賴模板的DNA聚合酶，可以催化脫氧核苷酸結(jié)合到斷裂DNA分子的3'-OH末端，且突出、凹陷或平滑末端的單鏈或雙鏈DNA分子均可作為其底物。細(xì)胞凋亡發(fā)生時(shí)，相關(guān)DNA內(nèi)切酶會(huì)被激活，從而切斷核小體間的DNA。在細(xì)胞凋亡晚期，DNA會(huì)被降解為180-200 bp的片段。 TdT介導(dǎo)的dUTP末端標(biāo)記法（TdT-mediated dUTP Nick-End Labeling，TUNEL）是在TdT的催化下將熒光素標(biāo)記的dUTP（fluorescein-dUTP）與斷裂DNA暴露的3'-OH聚合，延伸后的DNA可通過熒光顯微鏡或流式細(xì)胞儀進(jìn)行檢測。本產(chǎn)品適用于石蠟組織切片、冰凍組織切片、細(xì)胞爬片、細(xì)胞涂片以及細(xì)胞懸液。
